ABSORÇÃO DE NUTRIENTES PORVEGETAÇÃO DO SAPAL
A vegetação nas zonas de transição entre o sistema terrestre e aquático
desempenham um papel importante no controlo da entrada de materiais
nos sistemas aquáticos, nomeadamente nutrientes que, se em excesso,
podem promover “blooms” de algas, degradação da qualidade da água e
eutrofização. Alterações nas margens dos rios e ribeiras causadas por
práticas agrícolas, ou diminuição das zonas de sapal como consequência
da urbanização excessiva ou de alterações na hidrodinâmica dos
sistemas (p.ex. provocadas por barragens) podem reduzir a densidade e
ameaçar a presença desta vegetação. A reconstrução de ecotonos, como
zonas de sapal, é uma medida de gestão ambiental com potencialidade
de ser usada sempre que se pretender reduzir a entrada de
contaminantes nos sistemas aquáticos e desse modo controlar a
qualidade da água.

COMO FUNCIONA A FITOREMEDIAÇÃO?
Plant roots take contaminants from the ground into the
"body" of the plant. The plant root zone is referred to as the
rhizosphere, this is where the action occurs. This soil
supports large populations of diverse microorganisms. This
is due to chemicals exuded by plant roots which provide
carbon and energy for microbial growth. This combination of
plants and microorganisms appears to increase the
biodegradation of compounds.

PROCEDIMENTOS PRÁTICOS:
1 – lavar a vegetação e remover sedimento
2 – pesar cada conjunto e apontar valores
3 – colocar num copo cada conjunto separadamente e colocar etiqueta
4 – preparar solução com concentrações de fosfatos e nitratos
5 – medir as concentrações no fotómetro (seguir cuidadosamente as
indicações relativas à quantidade e tipo de reagentes a utilizar). Estas
medições constituem os valores T0.
6 – anotar os valores medidos
7 – colocar os frascos com a vegetação junto à janela para permitir o
fotoperíodo normal.

APLICAÇÃO PRÁTICA
BASE TEÓRICAIf freshwater mussels form a large component of the benthic biomass ofrivers (Mann, 1964), they may be expected to play an important role in riverfunctioning through their filtering. Invasive freshwater bivalves, such as D. polymorpha and C. fluminea, have had large impacts on river ecosystems (Phelps, 1994; Caraco et al., 1997). Quantifying the effect of freshwater mussels isimportant, because mussels are routinely (but accidentally) removed during riverdredging and weed-cutting operations (McIvor, 1999; Aldridge, 2000), and are often negatively affected by other forms of river management, such as the buildingof dams (Williams et al., 1993). If mussels are found to play important roles in waterpurification and river function, it will encourage managers to use ‘mussel-friendly’approaches to river management. This will benefit both the mussels themselves, many of which are considered a conservation priority (Willing, 1997; Neves, 1999), and will also help maintain healthy river function. In order to assess the magnitude of mussel filtration in natural waterbodies, it is firstnecessary to measure the filtration rates of individual mussels. These can then besummed over populations to estimate the population filtration rate of musselsin their natural environment. The filtration rate of a mussel is defined as the volume of water which passes through the mussel’s gills per unit time.

- Colocar cada grupo de bivalves em recipientes contendo 200 ml água do habitat natural da espécie;
-Um recipiente controlo sem bivalves será utilizado para corrigir a taxa de sedimentação;
- Medir a concentração de clorofila a “in vivo” inicial e após cada 30 min. durante uma hora. Para tal, retirar 5 ml de água com pipeta volumétrica, colocar numa cuvette de vidro para leitura.
-De forma a calibrar a concentração de clorofila a “in vivo, retirar amostras de água do branco inicial e filtrar através de filtros fibra de vidro Whatman GF-F, colocar em tubos de polipropileno e conservar a –80ºC, no escuro até procedimento de clorofila extractiva pelo método de Lorenzen (1967)
- Medir e pesar os bivalves (peso húmido), depois calcular peso seco (100ºC, 24h) e peso seco livre cinzas (450ºC, 3 h)

PROCEDIMENTOS PRÁTICOS
Taxa de excreção (TE):
- Colocar os bivalves em erlenmeyers contendo 200 ml de água
previamente saturada em oxigénio e filtrada por filtros Millipore (0,2 µm);
- Colocar dois erlenmeyers adicionais (controlo) sem bivalves para
detectar variações da concentração de amónia.
- Após 45 min , retirar 10 ml de água de cada um dos erlenmeyers e
determinar a concentração de amónia
PROCEDIMENTOS PRÁTICOS
Taxa de respiração
-Os bivalves serão transferidos para recipientes acrílicos fechados de volume conhecido (~780ml);
- A concentração de oxigénio será medida em intervalos de tempo regulares com uma sonda de oxigénio durante aproximadamente 1 hora, antes que a concentração diminua abaixo dos 30% do valor inicial de controlo;
- Um recipiente control sem bivalves será utilizado para corrigir respiração bacteriana, electrodos,etc.
